Specifically detects DNA polymerase delta p125 Clone: 11E10.1 in Human samples, and it is validated for Western BlottingDNA polymerase delta catalytic subunit (EC 2.7.7.7; UniProt P28340; also known as CDC2 homolog, DNA polymerase subunit delta p125) is encoded by the POLD1 (also known as CDC2, MDPL, POLD, CRCS10) gene (Gene ID 5424) in human. DNA replication in eukaryotes involves multiple enzymes including the B family polymerases Pol α, Pol δ, and Pol ?. While Pol α initiates DNA synthesis, Pol δ and Pol ? replace Pol α after primer extension and perform the bulk of DNA replication. Pol δ and Pol ? are heterotetramers in higher eukaryotes comprised of POLD1 and POLE as the respective catalytic subunit, as well as accessory subunits (POLD2/3/4 and POLE2/3/4) that interact with cofactors such as Proliferating Cell Nuclear Antigen (PCNA). Both POLD1 and POLE contain an exonuclease domain (a.a. 268-471 of POLE and 304-517 of POLD1) that proof-reading capability during DNA replication. In addition to DNA synthesis, Pol ? and Pol δ are believed to participate in chromosomal DNA repairs, including nucleotide excision repair (NER), ismatch repair (MMR) and repair of double strand breaks (DSBR). Germline exonuclease domain mutations (EDMs) in POLD1 and POLE cause polymerase proofreading-associated polyposis (PPAP). Tumors with EDMs exhibits dramatically increased base substitutions.
