The wells are washed and a mixture of biotinylated polyclonal anti-human C4a antibody and streptavidin-horseradish peroxidase is added, producing an antibody-antigen-antibody “sandwich”. The wells are again washed and substrate solution is added, which produces a blue color in direct proportion to the amount of C4a-desArg present in initial sample.
