Wells are washed and Streptavidin-horseradish peroxidase conjugate mixed with biotinylated anti-human IL-4 antibody is added, producing antibody-antigen-antibody “sandwich”. Wells are again washed and TMB substrate solution is added, which produces blue color in direct proportion to amount of IL-4 present in initial sample.
