The 5H4 antibody monoclonal antibody specifically binds to mouse CD122. CD122 is a 70-85 kDa type I transmembrane glycoprotein of the hematopoietin receptor superfamily. CD122 is also known as IL-2 Receptor beta chain (IL-2R β ) or IL-15 Receptor beta chain (IL-15R β ) as it helps form signaling receptor complexes for Interleukin-2 (IL-2) or IL-15, respectively. CD122 can combine with either CD132 (γc) alone or CD132 plus CD25 (IL-2Rα) to form inte rmediate or high-affinity IL-2 Receptor complexes, respectively. The IL-2R β chain is constitutively expressed on NK cells and at lower levels on some thymocytes, T cells, B cells, monocytes, and macrophages. IL-2R β chain expression is up regulated by IL-2. The immunogen used to generate this hybridoma was rat myel oma cells transfected with a truncated IL-2R β cDNA. The antibody was conjugated to BD Horizon BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407-nm and Em Max at 421-nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50-nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates.
