The 1A4 monoclonal antibody specifically binds to CD141. CD141 is a 75 kDa, single chain, type I membrane glycoprotein referred to as thrombomodulin and fetomodulin. CD141 is expressed on endothelial cells, smooth muscle cells, epithelial cells, synovial lining cells, and keratinocytes. It is also found on megakaryocytes, dendritic cells, peripheral blood moncytes, and neutrophils, but not on lymphocytes. Reports indicate that CD141 plays an important role in Protein C activation and the initiation of the Protein C anticoagulant pathway. Thrombin loses its procagulant function when it binds to CD141, and the CD141/thrombin complex is able to activate Protein C. The antibody was conjugated to BD Horizon BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407-nm and Em Max at 421-nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50-nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates.
