The csk gene encodes a protein kinase that phosphorylates several Src family kinases including p60(c-src), p59(fyn), p53/56(lyn), p56(lck), and p62(yes). This phosphorylation occurs specifically at the carboxy-terminal tyrosine, Y527. Csk (c-src kinase) was identified based on its ability to phosphorylate exogenous Src in mammalian src-deficient cells. Establishment of a mouse csk- cell line revealed that phosphorylation of Y527 in Src was reduced 20-50%. This correlated with an increase of Src kinase activity, thus indicating Csk is a negative regulator of src. Csk-deficient cells also show an elevation in the kinase activity of p59(fyn) and p53/56(lyn).
