Specifications HiGlutaXL™ medium contains the stabilized dipeptide form of L-glutamine, L-alanyl-L-glutamine. HiGlutaXL™ medium offers several advantages over the conventional glutamine containing media. Dipeptide form prevents the intramolecular cyclization reaction, thus preventing toxic build-up of ammonia. L-alanyl-Lglutamine incorporates L-alanine that protects the alpha amino acid group. Aminopeptidases within the cell break the dipeptide, gradually releasing both L-glutamine and L-alanine for use by the cell. The gradual release of Lglutamine obviates the need to supplement L-glutamine frequently and allows liquid media to be stored at 4°C for longer periods.C985F60, C985F61 and C985F62 is HiGlutaXL™ DMEM/Nutrient Mixture F-12 Ham with L-alanyl-L-glutamine, sodium bicarbonate, 15mM HEPES buffer and trace elements. HEPES, a zwitterionic buffer having a pKa of 7.3 at 37ºC prevents the initial rise in pH that tends to occur at the initiation of a culture and increases the buffering capacity of the medium. Users are advised to review the literature for recommendations regarding medium supplementation and physiological growth requirements specific for different cell lines.Storage and Shelf Life Store at 2-8ºC away from bright light. Shelf life is 18 months. Use before expiry date given on the product label.CompositionIngredientsmg/LInorganic SaltsAmmonium metavanadate0.00058Ammonium molybdate tetrahydrate0.00618Calcium chloride dihydrate154.500Copper sulphate pentahydrate0.0013Disodium hydrogen phosphate71.020Ferric nitrate nonahydrate0.050Ferrous sulphate heptahydrate0.417Magnesium chloride hexahydrate61.200Magnesium sulphate anhydrous48.840Manganese sulphate0.000151Nickel chloride0.00012Sodium bicarbonate1200.000Potassium chloride311.800Sodium chloride6996.000Sodium dihydrogen phosphate54.300Sodium metasillicate nonahydrate0.0142Sodium selenite0.00519Stannous chloride dihydrate0.00011Zinc sulphate heptahydrate0.432Amino AcidsGlycine18.750L-Alanine4.450L-Arginine hydrochloride147.500L-Asparagine monohydrate7.500L-Aspartic acid6.650L-Cysteine dihydrochloride17.560L-Cystine hydrochloride monohydrate31.290L-Glutamic acid7.350L-Alanyl-L-glutamine543.000L-Histidine hydrochloride monohydrate31.480L-Isoleucine54.470L-Leucine59.050L-Lysine hydrochloride91.250L-Methionine17.240L-Phenylalanine35.480L-Proline17.250L-Serine26.250L-Threonine53.450L-Tryptophan9.020L-Tyrosine disodium salt48.100L-Valine52.850VitaminsCholine chloride8.980D-Biotin0.0035D-Ca-Pantothenate2.240Folic acid2.660Niacinamide2.020Pyridoxal hydrochloride2.000Pyridoxine hydrochloride0.031Riboflavin0.219Thiamine hydrochloride2.170Vitamin B120.680myo-Inositol12.600OthersD-Glucose3151.000DL-Thioctic acid0.105HEPES buffer3574.500Hypoxanthine2.400Linoleic acid0.042Phenol red sodium salt8.630Putrescine hydrochloride0.081Sodium pyruvate110.000Thymidine0.365Quality ControlAppearanceRed colored, clear solutionpH7.00 -7.60Osmolality in mOsm/Kg H2O300.00 -340.00SterilityNo bacterial or fungal growth is observed after 14 days of incubation, as per USP specification.Cultural ResponseThe growth promotion capacity of the medium is assessed qualitatively by analyzing the cells for the morphology and quantitatively by estimating the cell counts and comparing it with a control medium through minimum three subcultures.Endotoxin ContentNMT 5EU/ml
