The Human Insulin-like Growth Factor Binding Protein 3 (Hu IGFBP3) ELISA quantitates Hu IGFBP3 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu IGFBP3. Principle of the method The Human IGFBP3 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. See manual for more information on validation.Insulin like growth factor binding protein 3 (IGFBP3) is a member of the superfamily of insulin like growth factor (IGF) binding proteins which include six high affinity IGF binding proteins (IGFBP) and at least four lowaffinity binding proteins referred to as IGFBP related proteins (IGFBPrP). The IGFBP members are cysteine rich proteins with conserved cysteine residues clustered in the amino terminal and the carboxy terminal regions of the molecule. The cDNA sequence encoding the mature human IGFBP3 is fused to the signal peptide of CD33. The 265 amino acid residue recombinant mature human IGFBP3 has a calculated molecular mass of approximately 29 kDa. Due to glycosylation, the recombinant protein migrates as 41 kDa in SDS-PAGE.
