The Human Interleukin 18 Receptor Beta (Hu IL-18RB) (IL-18RAP) ELISA quantitates Hu IL-18RB in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu IL-18RB. Principle of the method The Human IL-18RB solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. IL-18 receptor (IL-18R) is a type I membrane protein present in lung, leukocytes, spleen, liver, thymus, prostate, small intestine, colon, placenta and heart, absent from brain, skeletal muscle, pancreas and kidney. IL-18R is present in Hodgkin's disease cell lines, and does not bind IL-1α or IL-1β. The association of IL-18 to IL-18R leads to activation of NFκB. At present two subunits of IL-18R have been characterized: IL-18Rα and IL-18Rβ. IL-18Rα has been described as the ligand-binding chain and IL-18Rβ as the signal-transduction chain.
