The Human Interleukin 5 Receptor Alpha/CD125 (Hu IL-5RA) ELISA quantitates Hu IL-5RA in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu IL-5RA. Principle of the method The Human IL-5RA solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. See manual for more information on validation.Interleukin 5 (IL-5) is a soluble T cell-derived factor, also known as T cellreplacing factor (TRF), that induces B cell and eosinophil growth and differentiation. IL-5 exerts its biological activity through the IL-5 receptor (IL-5R),which is composed of two chains: an α chain that binds IL-5 with low affinity and a β chain that does not bind IL-5, but together with the IL-5 α chain,constitutes the high affinity IL-5 receptor. The cytoplasmic domain of both the α and β chains is essential for signal transduction. Specifically, the membrane-proximal proline-rich sequence of the ytoplasmic domain of the IL-5R receptor α subunit, IL-5Rα, is critical for the IL-5 induced proliferative response, expression of nuclear proto-oncogenes and tyrosine phosphorylation of cellular proteins, such as JAK1 and JAK2.
