The Human Kallikrein 6 (Hu KLK6) ELISA quantitates Hu KLK6 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu KLK6. Principle of the method The Human KLK6 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Kallikrein 6, or KLK6, shows activity against amyloid precursor protein, myelin basic protein, gelatin, casein and extracellular matrix proteins such as fibronectin, laminin, vitronectin and collagen. It degrades alpha-synuclein and prevents its polymerization, indicating that it may be involved in the pathogenesis of Parkinson disease and other synucleinopathies. KLK6 may be involved in regulation of axon outgrowth following spinal cord injury. KLK6 has also been shown to promote cell proliferation, migration, and invasion and is upregulated in several types of cancers including colon and ovarian cancers.
