The Human Programmed T cell Death 1 (Hu PD-1) ELISA quantitates Hu PD-1 in human serum, plasma or cell culture medium. The assay will exclusively recognize both natural and recombinant H PD-1. Principle of the method The Human PD-1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. PD-1 can bind two different but related ligands, PDL-1 and PDL-2. Upon binding to either of these ligands, signals generated by PD-1 inhibit the activation of the immune response in the absence of @@@danger signals@@@ such as lipopolysaccharide (LPS) associated with bacteria or pathogens. Evidence for this is seen in PD1-null mice who exhibit hyper-activated immune systems and autoimmune diseases. Further, expression of this gene in mice is induced in the thymus when anti-CD3 antibodies are injected and large numbers of thymocytes undergo apoptosis. Mice deficient for this gene bred on a BALB/c background developed dilated cardiomyopathy and died from congestive heart failure.
