The Human Siglec-9 ELISA quantitates Hu Siglec-9 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu Siglec-9. Principle of the method The Human Siglec-9 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. See manual for more information on validation.Siglec9 (Sialic acid-binding Ig-like lectin 9) is single-pass type I membrane protein and belongs to immunoglobulin super family and contains two Ig-like C2 type (immunoglobulin-like) domains and one Ig-like V-type (immunoglobulin-like) domain. It is reported that Siglec9 recognizes sialic acid in alpha2, 3-or alpha2, 6-glycosidic linkage to galactose. Using a specific antibody, Siglec9 was found to be expressed at high or intermediate levels by monocytes, neutrophils, and a minor population of CD16 , CD56- cells. Siglec9 also acts as an inhibitory receptor for MUC16 that attenuates T- and NK cell function. Recent reports suggest that Siglec9 expressed on DCs is involved in immuno regulation through ligation with mucins in an epithelial cancer patient.
