Stat1 EMSA (Electrophoretic-Mobility Shift Assay) kit is used for monitoring Stat1 (Signal transducer and activator of transcription 1) DNA binding activities and activation. Upon activation, Stat1 can recognize and interact with the unique DNA binding sequence and form protein/DNA complex, which can be separated from free DNA due to the differences in their electrophoretic mobility in native (non-denaturing) polyacrylamide gels. When one TF is activated, it binds to a biotin-labeled probe, which is designed based on the consensus binding sequence of the TF. When the formed complex is subjected to electrophoresis, it runs slower than the free probe, and can be easily distinguished. Two or more samples can be compared and differences can be determined by the density of the shifted band.
